Research Focus
Visualizing antibody responses to viral vaccines or infection using electron microscopy
My research focuses on understanding the atomic level interactions between antibodies induced by vaccination or natural infection, and viral glycoproteins. Cryo-EM methods are used to characterize isolated, recombinant monoclonal antibodies, with a focus on HIV vaccine research. In addition, I use electron microscopy to image and epitope map serum polyclonal antibodies in complex with vaccine immunogens, a technique named EMPEM. Combining high-resolution cryo-EM with EMPEM (cryoEMPEM), we can understand the specific amino acids of the antigen contacted by polyclonal antibodies and also predict possible sequences of the antibodies themselves, accelerating vaccine design and antibody discovery.
