MicroRNAs (miRNAs), discovered in 1993, are evolutionarily conserved, small noncoding, gene-silencing, 22-nt long RNAs found in Eukaryotes. Animal miRNAs regulate gene expression at the level of translation by using imperfect target complementarity with cognate mRNA in the 3' untranslated regions (UTRs), resulting in translational repression and/or degradation of the target transcript. Regulatory roles of miRNAs have been implicated in diverse developmental, physiological and pathogenic events. However, the role and function of miRNAs in regulating human T and B lymphocyte activation in models for acute, antigen-driven immune responses has not been studied. We are interested in studying the role that differentially expressed miRNAs play in regulating human T and B lymphocyte activation and if they do so by targeting highly differentially expressed and/or alternative spliced genes. Our study encompasses a systematic genome-wide analysis of alternative splicing as a function of activation in human lymphocytes while also inclusively analyzing genome-wide differential gene and miRNA expression.
