Leica TCS SP5 Two-Photon Microscope  (Room IMM309A)


The Leica SP5 multiphoton system is equipped with a pulsed Mai Tai laser (710 – 990 nm) and sensitive detectors and allows for real-time, intravital imaging in live animals or whole organs.  Because of the ability of the longer wavelength laser to penetrate deeper into tissue (several hundred microns), it is useful for imaging whole mouse organs such as lymph nodes, lens, skin, etc.  In addition to the 2-photon laser, the microscope is also equipped with 3 visible lasers that generate 5 individually controlled laser lines at 458, 488, 514, 543, and 633 nanometers.  A long working distance water immersion objective with high numerical aperture (20x, 0.95 NA, Olympus) is used for both excitation and collection of the backscattered light.  The microscope is equipped with 4 high sensitivity non-descanned detectors (NDD) that are separated into 4 channels (DAPI-FITC/Alexa568 or Rhodamine/Alexa 594 or Texas red/ Alexa 647 or Cy5) using three long pass dichroic mirrors at 562 nm, 593 nm and 665 nm. In addition it has 5 descanned detectors.
